Isis Reports Interim Phase 2 Data on ISIS-APOCIII Rx as a Single Agent in Patients With Very High to Severely High Triglycerides

CARLSBAD, Calif., Aug. 31, 2013 /PRNewswire/ -- Isis Pharmaceuticals, Inc. (NASDAQ: ISIS) announced interim data from an ongoing Phase 2 study of ISIS-APOCIIIRx as a monotherapy in patients with very high to severely high triglycerides.  These data were presented today by Dr. Daniel Gaudet at a PACE session occurring concurrently with the European Society of Cardiology in Amsterdam.  In this study, patients treated with ISIS-APOCIIIRx achieved statistically significant mean reductions of up to 79 percent in apolipoprotein C-III (apoC-III) and up to 75 percent in triglycerides.  In addition, patients treated with ISIS-APOCIIIRx achieved statistically significant mean increases of up to 57 percent in high-density lipoprotein cholesterol (HDL-C), the 'good' cholesterol.  Patients also achieved up to 89 percent mean reduction in apoC-III-associated very low-density lipoprotein (VLDL) particles.

"Patients with very high levels of triglycerides experience a significant number of health problems, including type 2 diabetes, obesity, cardiovascular disease and, for patients with severely elevated triglycerides, pancreatitis.  For these patients, there are very limited treatment options and what is needed is a much more effective drug that can be used as a single agent or in combination with other triglyceride-lowering agents," said Daniel Gaudet, M.D., Ph.D., from the department of medicine, University of Montreal and scientific director, Genome Quebec Biobank Technology Center.  "The comprehensive Phase 2 data reported this summer demonstrate that ISIS-APOCIIIRx has the potential to fill this need."

The monotherapy portion of the ongoing Phase 2 study of ISIS-APOCIIIRx is a double-blind, randomized, placebo-controlled 13-week study designed to assess the safety and activity of ISIS-APOCIIIRx in patients with very high to severely high triglyceride levels (between 440 and 2,000 mg/dL).  The data reported today is an interim analysis of 28 patients who completed 13 weeks of treatment with ISIS-APOCIIIRx or placebo.  In this study, patients treated with ISIS-APOCIIIRx experienced dose-dependent, robust and prolonged, mean percent reductions from baseline in apoC-III, triglycerides and apoC-III-associated VLDL particles.  Furthermore, these patients demonstrated a rapid, prolonged and statistically significant mean percent increase from baseline in HDL-C.

In the ongoing study, patients received 100 mg, 200 mg or 300 mg dose of ISIS-APOCIIIRx, or placebo via weekly subcutaneous injections.  In this interim analysis, patients had an average fasting triglyceride level of 602 mg/dL with incoming triglyceride levels up to 1,822 mg/dL.  In this study ISIS-APOCIIIRx was found to be generally safe and well tolerated.  The most common adverse event (AE) was injection site reactions, which were infrequent, predominantly mild and typically resolved rapidly.  There were no flu-like symptoms, no treatment-related elevations of liver enzymes greater than three times upper limit of normal, no abnormalities in renal function and no clinically meaningful changes in other laboratory values.

We have now demonstrated that ISIS-APOCIIIRx is equally effective in patients with high to severely high triglycerides as well as in patients with high triglycerides and type 2 diabetes.  We have demonstrated that ISIS-APOCIIIRx can work equally well as a single agent and in combination with fibrates to produce significant reductions in apoC-III and triglycerides, and increases in HDL-C.  In addition, the positive effect of ISIS-APOCIIIRx treatment on lipid parameters, improvements in glucose control and trends toward improvements in insulin sensitivity, suggest that ISIS-APOCIIIRx could have a very attractive therapeutic profile for patients with severely high triglycerides, who often also have diabetes or metabolic syndrome," said Richard Geary, Ph.D., senior vice president of development at Isis.  "We look forward to discussing our Phase 3 plans with regulators and moving rapidly into a Phase 3 program next year in patients with severely high triglycerides."

ISIS-APOCIIIRx is an antisense drug intended to treat patients with severely high triglycerides either as a single agent or in combination with other triglyceride-lowering agents.  ISIS-APOCIIIRx targets apoC-III, a gene produced in the liver that plays a central role in the regulation of serum triglycerides.  Humans who do not produce apoC-III have lower levels of triglycerides and lower instances of cardiovascular disease.  In clinical studies, patients with lower levels of apoC-III and triglycerides exhibit lower cardiovascular event rates.  Humans with elevated levels of apoC-III have increased dyslipidemia associated with multiple metabolic abnormalities, such as insulin resistance and/or metabolic syndrome.  In addition, the prevalence of type 2 diabetes is increased in patients with elevated triglycerides.

Dear Tesco, please stop encouraging me to buy things that I really don’t need! I have fallen prey to your voucher system more times than I can count and now own about seventy-two Fissler pans, a set of Pyrex oven-to-hob-to-freezer-to-whatever dishes and enough cutlery to serve an army with!

I simply cannot resist something that, on the face of things, seems like a bargain. Take these Fissler pans for example: the stockpot, with ten vouchers, cost me £29.99. In Harrods (I checked) it was just under £200. WHAT AN IMMENSE SAVING! I thought, and then proceeded to buy every single pot and pan that was on offer. Going backwards and forwards to Tesco, collecting the vouchers (one per £20 spent, you work it out!) and feeling pleased as punch as I added pan after pan to my collection.

Pleased as punch until Mr AMR worked out how much I had been spending. I could have bought a whole set of kitchenware for the amount I’ve invested in my Fissler pans! I could have bought an entire bloody kitchen! Can someone please remind me, next time that Tesco entice me with an offer, that I do NOT need any more kitchen stuff? Printer ink, yes. A new linen basket – yes. Pans, no.

Here I paired one coat of Boyfriend Material with a pale pink (Revlon Brilliant Strength Nail Enamel in Allure), and it’s just glitter heaven. In the bottle, this polish leans towards a green shade, so I didn’t know what to expect once it was on the nail, but it is B-E-A-U-T-I-F-U-L! It’s packed with tiny green and gold glitters with flecks of silver throughout, which make it sparkly but in a subtle way. It can easily be worn all day, every day. Which is exactly how I think I might wear it. If this polish were a boy, he would definitely be boyfriend material.

Big Red reminds me of Dorothy’s slippers—it’s a true ruby red with what looks like little ruby glitters suspended throughout. This one is a jelly (meaning it’s not totally opaque), which I love because as you layer the polish, the glitter doesn’t get lost—it just seems suspended between the layers, adding tons of depth to your mani. This was two easy coats, but be warned: the scent on this polish is beyond strong. I felt like I was in a nail salon sitting next to someone getting acrylics (don’t ever get acrylics, girls!) and I really couldn’t stand it. The good news is I was assured the scent was simply due to the polish being made and bottled right away—it should fade quickly. Here’s hoping, because this colour is too good not to wear!

Spanky Pants is a medium grey packed with lime, neon pink, black and white glitters. And it’s all kinds of great. It took me three thin coats to get opaque, but you could definitely do with two if you have shorter nails and don’t mind seeing your nail line peek through. I loved the colour combo on this one; anything neon paired with grey is sure to be a hit in my books.

Spirit Fingers

Spirit Fingers (you knew this name would be here!) reminds me of the good ol’ US of A: it’s red, white and blue all over. Three coats will give you full opacity, but the polish can get a little thick on the nail. The milky white base isn’t super-opaque, so this polish has tons of depth, too. Just look at all those glitters peeking through—S.W.O.O.N!

B-E Agressive

B-E Aggressive is another milky white base, but this time with square black, blue and pink glitters plus a very fine sparkle running throughout. Though it’s similar to Spirit Fingers, I definitely think each is unique in its own way. This applied really well (I did three coats plus top coat). Love!

My favourite of all six was the namesake of the collection, Cheer-ocracy. It’s a glitter topcoat, so naturally, I was immediately sold. I decided to try it over a peachy base, Sally Hansen Complete Salon Manicure Nail Polish in Peach of Cake, and I couldn’t get over how much I loved it. It just… makes me want to cheer! And it totally reminds me of the pom-poms cheerleaders flail around. This was just one coat and could definitely be built up with another for more glittery goodness. I am a fan.

I’m really loving indie nail brands at the moment—they just deliver something completely different. Plus, the part of me that secretly wished she cheered at an all-American high school, Bring It On-style, is overjoyed right now with this collection.

Let’s make one thing clear, I don’t ever get excited about cleaning or cleaning products. It takes me all day to clean the bathroom (probably because there is so much stuff in there). Our kitchen always looks like vegetable massacre scene because I try to cook from scratch as much as possible. I’ve always cleaned everything with Domestos spray bleach and James likes to use water and vinegar in spray bottle. This means that the house ends smelling like a chippy or hospital – neither of which is ideal. When I was offered to try some Method products I was a bit sceptical because I’ve used ‘environmentally friendly’ cleaning products before but they just didn’t seem up to the job. Well now I’m eating my words!

Method Daily Kitchen Cleaner is fooking amazing and here’s why:

  • Cleans all manner of grease and sticky surfaces
  • Non-toxic made from plant ingredients
  • Scent is fresh and not overpowering
  • Doesn’t have a ‘middle class’ price tag (£3 for 828ml bottle)

As I share my house with Trenty my 10-year-old house panther, it’s important that I don’t use anything with harsh chemicals on places he might go. He’s a good boy and never jumps on the kitchen counter tops, but he does like to lay on my desk near the window and uses a cat flap. This cleaned his cat flap beautifully (it was filthy) and the surface of my desk which he probably gets dirty again with all the stuff attached to his fur from outside. Either way I know he won’t be harmed by Method.

There are loads of other cleaning and household products that Method do and they really are worth a look if you are looking for an alternative to traditional household chemicals. Method products are available on their website, supermarket & diy stores, and green product websites.

Now it’s very rare that I deviate from talking about beauty products on the blog but I just had to get this off my chest. Now I think a lot of people knew that the beautiful CB wouldn’t return to play Batman even though most fans including myself were secretly praying that he would. But there was also a little part of me that said the trilogy should close thus leaving another Batman reboot far into the future like when I’m 90 or something. Having learned yesterday that Ben (So boring I’d rather watch paint dry, have a cavity filled, clean the toilet, and muck our horse stables) Affleck was replacing my Christian as Batman I just felt sick.

Okay I know there is a lot of sh*t going on the world that is more important but for Entertainment purposes hear me out. The reason I love Mr. Bale is because he’s a versatile actor who doesn’t care how he looks and is willing to be ugly for a film role. Christian Bale isn’t your typical Hollywood actor, he’ll never be on Celebrity Big Brother or be photographed with lots of honeyz. Yes some of his films have been rubbish (Equilibrium anyone?) but when he’s good he’s really good (The Fighter, 3.10 to Yuma, and of course American Psycho). He’s equally comfortable doing big budget and and independent films and seems like an all round decent married man with a child. And he does a fantastic American accent that is totally believable, so much so that his ‘normal’ British accent sounds a bit strange.

I could go on for days, as a matter of fact I’m sure I could do an entire conference about the man but I shall leave you with a favourite scene from American Psycho which is totally relevant to Skin Scrubs – Enjoy!

Verizon Wireless rolls Cell on Wheels (COW) into Plumas County for firefighters

What:

Verizon Wireless responded to a request from local authorities for additional network coverage and capacity to support emergency personnel battling the Plumas Lightning Complex Fires. The additional temporary network equipment, located in Taylorsville, CA, will enhance wireless voice and data coverage for emergency personnel and others.




Boosting coverage and capacity to existing cell sites in the area also enhances wireless coverage so that emergency response teams and other Verizon Wireless customers can make more calls and send more data transmissions faster.




Verizon Wireless has also provided wireless phones, tablets, and 4G LTE Jetpacks to fire command crews to enhance voice and data communications at the base camp.

This routine takes things right back to the skincare basics. For many of you, it may be slightly too obvious, a bit “common sense-y” and for that I’m sorry, but I just felt that I wanted to get some simple skincare steps recorded on video. The importance of proper cleansing, mainly, I suppose, but then a little note about serums and treatments and where they fit in to a daily beauty routine. Because I do feel that a lot of weight is afforded to moisturisers - always be sure to use a good moisturiser! - but that many moisturisers are quite limited in performance in comparison to serums and things like BHA treatments and overnight peels. I get much more out of my Liquid Gold or Advanced Night Repair II than I do from only using a run-of-the-mill moisturiser; the treatment serums seem to really get to work and change the appearance of my skin, whereas the moisturisers mostly just seem to sit there and read the papers. OK, I exaggerate – there are some brilliant, brilliant moisturisers out there, and they are an entirely necessary part of the routine – but I suppose that I want to stress the importance of a good treatment or serum. The textures are usually finer so that they can get to work more easily on the skin and they normally have quite concentrated ingredients. Take Liquid Gold, for example, for intense exfoliation and rejuvenation or Hydraluron for a massive shot of unadulterated moisture. You wouldn’t get quite the same effects with most face creams – they serve a different purpose. They do a great job of keeping the skin plump and juicy and soft, but as a rule, it’s the serum beneath that’ll be delivering the skincare goods.

I’ll repeat as a rule, because I know that you’ll come up with about three zillion examples where this isn’t true: again, I’m by no means saying that moisturisers are useless – you’ll see when you watch the video that they form an entire THIRD of my beauty routine! – I’m just trying to advocate serums and treatments as brilliant things. When it comes to addressing specific needs they are simply irreplaceable; BHAs or salicylic acid for helping with spots, glycolic or AHAs for keeping skin looking rejuvenated and fresh, treatments for dark spots, serums for hydration, potions for redness and sensitivity and tightness… There are moisturisers that contain these ingredients – many now are incredibly hi-tech – I just can’t see the harm in giving the skin a pre-moisturiser treat. Something that will do the business on whatever it is that you want to fix. And if there’s nothing that needs fixing then good for you! Though I still think that you can invest in your skin’s future and use a serum that will give a little extra protection during the day – something antioxidant and clever like the CE Ferulic serum from Skinceuticals (pricey but excellent) or the Skinesis Age Repair Serum from Sarah Chapman. None of this is gospel, obviously, this is just what I do. And yes, serums do tend to be pricier, but I’ve given a few affordable suggestions that I’ve tested over the past year – you can find a list of ideas at the bottom of this post.

A word on toners, because there were lots of comments on them underneath the Youtube video after I said that I didn’t count them as a step: I am in no way saying that I don’t agree with toners. I use a toner now and then, I just don’t see it as an absolutely crucial part of my routine. If I was a cotton-wool-and-lotion type of girl then I would no doubt follow up with a toner, but I do a thorough cleanse with a balm or cream cleanser and this, on its own, usually works well for me. My skin feels ultra-clean and balanced. The reason I don’t push toner as an essential part of my routine is that lots of people I know reach for the most alcoholic, skin-stripping toner they can find (my personal teenage favourite used to be a bright pink concoction from Clinique that made my eyes water) and then use this as a secondary cleanse. Not ideal. If you have a good one and you love it, go ahead – I quite like Lancome’s one here, and I don’t mind the occasional sweep of the Clarins exfoliating or my MD Formulations Anti-Oxidant spray, but I don’t go into panic-mode if I can’t find one in the cesspit of my beauty room.

I do a proper, full cleanse (as shown here in the video) at night. In the morning it’s usually a quick wash with a cleansing wash or I use cleansing waters. Mostly I’ll use cleanser and water though! It’s really important to do a thorough cleanse – if you need to do it twice to get off all the crap then so be it!

I still use the Clarisonic but mostly as part of a home facial or occasional deep cleanse. It’s too much for me to use every day. But if you get on well with it, use as much as you like.

I haven’t included exfoliants in this video because I don’t use scrubs, really, which is what most people think of when you say “exfoliation”. I prefer regular chemical exfoliation – I use Liquid Gold every two or three nights, but not so much in the summer as it does make the skin a little more sensitive to the sun.

Those with acne/oily skin may want to use some kind of exfoliant on a regular basis to keep pores clear and refined. BHAs/salicylic acid are great for exactly this. You’d more often than not use your exfoliating treatment or serum in the SERUM/TREATMENT step before moisturiser. Not always, but mostly. Paula’s Choice do great BHA products with different strengths.

Lots of the serums I use contain mild exfoliants – the Liquid Gold Rejuvenating, for example, and the new Chanel Weekend product. Anything with glycolic and lactic acids and other AHAs in will be exfoliating as they work. Great for pepping up the skin and making it fresh and clear. See below – serums!
